目的采用HPLC法测定黄毛翠雀花中乌头碱、新乌头碱和次乌头碱的含量。方法采用Aglient Eclipse XDB-C18色谱柱(150 mm×4.6 mm,5μm),流动相为甲醇-0.03%三乙胺(72∶28),流速1.0 mL·min-1,柱温25℃,检测波长235 nm。结果乌头碱、新乌头碱和次乌头碱的线性范围为0.14~0.84μg(r=0.9999)、0.206~1.236μg(r=0.9998)、0.18~1.8μg(r=0.9999);平均回收率分别为100.05%(RSD=0.24%)、101.42%(RSD=0.32%)、98.81%(RSD=0.27%)。结论所用方法操作简单,准确性、重复性好,可用于黄毛翠雀花中乌头类物质的质量控制。
OBJECTIVE To determine the content of aconitine, mesaconitine and hypaconitine in Pterocarya stenptera by HPLC. Methods Aglient Eclipse XDB-C18 column (150 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of methanol-0.03% triethylamine at 72:28, the flow rate was 1.0 mL · min- 235 nm. Results The linear ranges of aconitine, mesaconitine and hypaconitine ranged from 0.14 to 0.84 μg (r = 0.9999), from 0.206 to 1.236 μg (r = 0.9998) and from 0.18 to 1.8 μg (r = 0.9999) Rates were 100.05% (RSD = 0.24%), 101.42% (RSD = 0.32%) and 98.81% (RSD = 0.27%), respectively. Conclusion The method is simple, accurate and reproducible. It can be used for the quality control of aconites in Pseudostellaria barbata.
