目的系统评价肿瘤坏死因子α诱导蛋白3相互作用蛋白1(TNFα-induced prote in 3-interacting protein 1,TNIP1)基因与系统性红斑狼疮(systemic lupus erythematosus,SLE)的相关性。方法计算机检索Pub Med、Web of Science和EMbase,查找关于TNIP1基因rs7708392或rs10036748位点多态性与SLE相关性的病例-对照研究,检索时间至2015年2月;由2名评价员按照纳入和排除标准独立进行文献筛选、资料提取和质量评价后,采用Stata 12.0软件进行Meta分析。结果共纳入12篇研究文献,其中关于TNIP1基因rs7708392位点多态性与SLE的病例-对照研究10个,关于TNIP1基因rs10036748位点多态性与SLE的病例-对照研究6个。Meta分析结果显示,病例组TNIP1基因rs7708392位点等位基因C频率高于对照组(OR=1.23,95%CI:1.12~1.34,P<0.001);病例组TNIP1基因rs10036748位点等位基因T频率高于对照组(OR=1.29,95%CI:1.24~1.34,P<0.001);按照种族分层分析结果显示,欧洲人群TNIP1基因rs7708392位点C:T的OR值为1.29(95%CI:1.23~1.35),亚洲人群C:T的OR=1.14(95%CI:0.92~1.42);亚洲人群TNIP1基因rs10036748位点T:C的OR为1.25(95%CI:1.18~1.33)。结论 TNIP1基因rs7708392位点C等位基因、rs10036748位点T等位基因与系统性红斑狼疮遗传易感性相关,TNIP1基因上述位点基因型有可能成为早期诊断系统性红斑狼疮的指标。
Objective To evaluate the relationship between TNFα-induced proteoin 3-interacting protein 1 (TNIP1) gene and systemic lupus erythematosus (SLE). METHODS: PubMed, Web of Science and EMbase were searched by computer to find a case-control study on the association of TNIP1 rs7708392 or rs10036748 polymorphism with SLE. The search time was up to February 2015. Two reviewers, Exclusion criteria Independent literature screening, data extraction and quality evaluation, the use of Stata 12.0 Meta-analysis software. Results A total of 12 research articles were included, of which 10 were case-control studies on rs7708392 polymorphism of TNIP1 gene and SLE, and 6 case-control studies on rs10036748 polymorphism of TNIP1 gene and SLE. Meta analysis showed that the frequency of allele C in rs7708392 of TNIP1 gene in case group was higher than that in control group (OR = 1.23, 95% CI: 1.12-1.34, P <0.001) (OR = 1.29,95% CI: 1.24-1.34, P <0.001). According to the ethnic stratification analysis, the OR of C: T of rs7708392 in TNIP1 gene of European population was 1.29 (95% CI : 1.23-1.35), OR = 1.14 (95% CI: 0.92-1.42) for C: T population in Asia and 1.25 (95% CI: 1.18-1.33) for T: C at rs10036748 locus in Asian population. Conclusion The allele C of rs7708392 and the allele rs10036748 of TNIP1 gene are associated with the genetic predisposition of systemic lupus erythematosus. The above-mentioned loci genotype of TNIP1 gene may be an index for early diagnosis of systemic lupus erythematosus.
