目的研究常用人工合成麝香吐纳麝香(AHTN)和佳乐麝香(HHCB)对129小鼠胚胎干细胞的发育毒性效应。方法利用MTT法检测两种麝香对胚胎干细胞的毒性效应,结合不同暴露浓度下麝香对胚胎干细胞体外分化为心肌细胞能力的抑制情况,判断受试物的发育毒性。结果 MTT实验表明,AHTN和HHCB对小鼠胚胎干细胞半数毒性浓度IC50分别为34和29μg/ml,而分化抑制实验表明这两种麝香对胚胎干细胞向心肌细胞的半数分化抑制浓度ID50分别为257和153μg/ml。结论人工麝香对胚胎发育具有弱发育毒性,对孕妇和儿童发育的影响需要在相关的发育毒性研究中给予重视。
Objective To investigate the developmental toxic effects of commonly used synthetic musk thumne (AHTN) and musk musk (HHCB) on 129 mouse embryonic stem cells. Methods MTT assay was used to examine the toxic effects of two musk on embryonic stem cells. Combined with the inhibitory effect of musk on the ability of embryonic stem cells to differentiate into cardiomyocytes in vitro under different exposure concentrations, the developmental toxicity of musk was evaluated. Results MTT assay showed that IC50 of AHTN and HHCB on mouse embryonic stem cells were 34 and 29 μg / ml, respectively. However, the inhibitory effect of these two musk on ICCs of cardiomyocytes was 257 and 153 μg / ml. Conclusion Artificial musk has weak developmental toxicity on embryo development, and its influence on the development of pregnant women and children needs to be paid more attention in the related developmental toxicity study.
