目的 观察干扰素γ(Interferon -γ ,IFN -γ)对兔耳创面愈合期伤口周边组织和肉芽组织中蛋白酪氨酸激酶(ProteinTyrosineKinase,PTK)活性以及伤口愈合时间的影响 ,探讨IFN -γ外用于皮肤创面的信号传导机制。 方法 在兔耳腹侧伤口外用IFN -γ,观察上皮化的时间并测定正常兔耳、伤后 1天、3天、6天和创面上皮化时 (12~ 14天 )组织中PTK活性变化。结果 周边和肉芽组织中PTK活性伤后均高于正常皮肤组织 (P <0 .0 1) ,周边组织伤后 6天即恢复正常水平 ,而肉芽组织在上皮化后仍然较高。IFN -γ可使伤口周边和肉芽组织的PTK活性升高 (P <0 .0 5或 0 .0 1)。IFN -γ可使伤口愈合延迟 1.2 3天 (P <0 .0 1)。结论 创伤愈合组织中PTK活性升高且与增生程度相关。PTK既介导生长刺激信号使细胞增殖和合成物质的功能增强 ,也介导IFN -γ的信号使愈合延迟。
Objective To investigate the effects of Interferon-γ (IFN-γ) on the activity of protein tyrosine kinase (PTK) in wounds and granulation tissue in rabbits ear wound healing and wound healing time. Signal transduction mechanism in skin wounds. Methods IFN-γ was applied to the ventral wound of rabbit ear to observe the time of epithelialization. The changes of PTK activity in normal rabbit ear, wounds at 1 day, 3 days, 6 days and wound epithelialization (12 ~ 14 days) were observed. Results Peripheral and granulation tissue PTK activity after injury were higher than normal skin tissue (P <0.01), the surrounding tissue 6 days after injury to return to normal levels, while the granulation tissue after epithelization is still high. IFN-γ increased the PTK activity in wounds and granulation tissue (P <0.05 or 0.01). IFN-γ delayed wound healing by 1.2 days (P <0.01). Conclusions PTK activity in wound healing tissue is elevated and correlates with the degree of proliferation. PTK mediates growth stimulation signals to enhance cell proliferation and the function of synthetic substances. It also delays the healing of IFN-γ signaling.
