目的 研究姜黄素对人类Burkitt淋巴瘤体外抗癌作用 ,探讨其抗癌作用的分子机制。方法 应用台盼蓝拒染法、MTT法、细胞周期分析法、TUNEL法及DNA片段化分析等方法 ,检测姜黄素对CA4 6细胞生长及凋亡的影响 ,并通过流式细胞术、RT PCR检测姜黄素对CA4 6细胞c myc、bcl 2、突变型p5 3、Fas蛋白和mRNA表达的影响。结果 (1)姜黄素抑制CA4 6细胞增殖呈时效及量效关系。(2 )姜黄素处理组CA4 6细胞周期发生变化 ,G0 /G1或G2 /M期细胞比例增多 ,S期细胞比例减少。 (3)姜黄素处理组CA4 6细胞发生凋亡 ,表现为TUNEL法可检测到细胞内呈棕色颗粒的凋亡细胞 ,DNA片段化分析可出现典型的梯状DNA条带。 (4)姜黄素处理组CA4 6细胞c myc、bcl 2、突变型p5 3蛋白和mRNA表达水平显著下降 ,而Fas蛋白和mRNA表达水平则上升。结论 姜黄素能够抑制CA4 6细胞增殖 ,并诱导其凋亡 ,推测可能是通过下调c myc、bcl 2、突变型p5 3,上调FasmRNA及蛋白的表达而起作用。
Objective To study the anticancer effect of curcumin on human Burkitt lymphoma in vitro and to explore the molecular mechanism of its anticancer effect. Methods The effects of curcumin on the growth and apoptosis of CA4 6 cells were detected by trypan blue exclusion method, MTT assay, cell cycle assay, TUNEL assay and DNA fragmentation analysis. Flow cytometry, RT PCR The effect of curcumin on the expression of c myc, bcl 2, mutant p5 3, Fas protein and mRNA in CA4 6 cells was detected. Results (1) Curcumin inhibited CA4 6 cell proliferation in a time-and dose-effect relationship. (2) Curcumin treatment group CA4 6 cell cycle changes, G0 / G1 or G2 / M phase cells increased, S phase cells decreased. (3) Apoptosis of CA4 6 cells was observed by curcumin treatment. TUNEL assay could detect apoptotic cells with brown granules in the cells, and typical ladder DNA bands appeared in DNA fragmentation analysis. (4) The expression of c-myc, bcl 2, mutant p5 3 protein and mRNA in CA4 6 cells decreased significantly while the expression of Fas protein and mRNA increased in curcumin treated group. Conclusion Curcumin can inhibit the proliferation of CA4 6 cells and induce its apoptosis. It is speculated that curcumin may play a role in down-regulation of c-myc, bcl-2, and mutant p5 3 by up-regulating the expression of Fas mRNA and protein.
