目的研究大鼠心肌梗死后心肌转化生长因子β1(TGF-β1)与Ⅰ型胶原表达,并测定大鼠血浆内TGF-β1的含量。方法结扎左冠状动脉建立大鼠心肌梗死模型,采用ELISA法测定心肌梗死不同时间内大鼠血浆内TGF-β1的含量;应用免疫荧光染色的方法,通过激光扫描共聚焦显微镜观察TGF-β1与Ⅰ型胶原表达的变化。结果大鼠心梗后1 d3、d5、d血浆内TGF-β1含量明显增加(P<0.01),呈时间依赖性。心梗后24 h心肌TGF-β1与Ⅰ型胶原表达与假手术组相比明显增强。结论心梗后心肌通过增加TGF-β1、Ⅰ型胶原的表达参与心室重构。
Objective To investigate the expression of transforming growth factor β1 (TGF-β1) and type Ⅰ collagen in rats after myocardial infarction and to determine the content of TGF-β1 in rat plasma. Methods The model of myocardial infarction was established by ligation of the left coronary artery in rats. The content of TGF-β1 in the plasma was determined by ELISA. The expression of TGF-β1 and TGF-β1 in the plasma were detected by laser scanning confocal microscopy Type collagen expression changes. Results The content of TGF-β1 in the plasma on 1 d3, d5 and d after myocardial infarction in rats increased significantly (P <0.01) in a time-dependent manner. The expression of TGF-β1 and type Ⅰ collagen in myocardium 24 h after myocardial infarction was significantly higher than that in sham operation group. Conclusions After myocardial infarction, myocardium participates in ventricular remodeling by increasing the expression of TGF-β1 and type Ⅰ collagen.
