为了研究银杏GRP 1 8启动子在木本植物中的调控制特性 ,用从银杏细胞壁形成及纤维素生物合成密切相关的 ,并定位于形成层木质部维管组织细胞表达的富甘氨酸蛋白质基因 (GRP 1.8)的启动子与GUS报告基因构建的表达载体转化烟草 ,经过Southernblotting鉴定 ,得到一批转化再生植株 .经GUS组织化学检测 ,发现转基因烟草的茎木质部呈现GUS染色阳性 ,初步证明银杏GRP 1 8基因启动子确有韧皮部表达特性 ,可介导GUS基因在转基因烟草木质部特异性表达
In order to study the regulatory properties of Ginkgo biloba GRP 1 8 promoter in woody plants, we used the glycine-rich protein gene (GRP), which is closely related to ginkgo cell wall formation and cellulose biosynthesis and located in the cambium of vascular tissues of xylem 1.8) promoter and GUS reporter gene were transformed into tobacco and identified by Southern blotting, a batch of transformed plants was obtained.GUS histochemical examination showed that the stem xylem of transgenic tobacco showed positive GUS staining, which initially proved that Ginkgo biloba GRP 1 8 Gene promoter does have phloem expression characteristics, can be mediated GUS gene specific expression in transgenic tobacco xylem
