目的研究金苓痛风舒微丸的抗炎作用及其作用机制。方法采用二甲苯致小鼠耳廓肿胀及角叉菜胶致大鼠足肿胀实验探索金苓痛风舒微丸的抗炎作用。采用腺嘌呤、乙胺丁醇灌胃及关节腔注射尿酸钠复合造模的方法复制大鼠痛风性关节炎模型。将大鼠随机分为正常组,模型组,别嘌醇组(0.036 g·kg-1),痛风定组(0.432g·kg-1),金苓痛风舒微丸高、中、低剂量组(37.8,18.9,9.45 g·kg-1),共7组,每组10只。采用酶联免疫法(ELISA)检测血清中前炎症因子肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)及白介素-8(IL-8)的表达。并观察关节滑膜组织病理形态学的改变。结果金苓痛风舒微丸可有效抑制小鼠耳廓炎症肿胀(P<0.05)及大鼠足肿胀(P<0.05,P<0.01)。显著抑制痛风性关节炎大鼠血清中TNF-α、IFN-γ、IL-8的表达(P<0.05,P<0.01),减轻大鼠关节滑膜的炎性细胞浸润、充血、肿胀和坏死(P<0.05,P<0.01)。结论金苓痛风舒微丸对急性炎症具有显著的抗炎作用,其对痛风性关节炎的抗炎机制可能是通过抑制前炎症因子TNF-α、IFN-γ、IL-8的表达,进而调控滑膜组织中炎症反应链,从而发挥抗炎作用。
Objective To study the anti-inflammatory effect and mechanism of Jinling Tongfeng Shuwei Pills. Methods The anti-inflammatory effect of Jinling Tongfengshuwei Pill was explored by xylene-induced auricle swelling and carrageenan-induced foot swelling in rats. The gouty arthritis model was reproduced by adenine, ethambutol intragastric and intraarticular injection of sodium urate. The rats were randomly divided into normal group, model group, allopurinol group (0.036 g · kg-1), Tongfengding group (0.432g · kg-1), Jinling Tongfengshuwei Wan high, medium and low dose group (37.8,18.9,9.45 g · kg-1), a total of 7 groups, 10 in each group. Serum levels of TNF-α, IFN-γ and IL-8 were detected by enzyme linked immunosorbent assay (ELISA). And observe the synovial pathological changes in the morphology. Results Jinling Tongfeng Shuwei Pill could effectively inhibit swelling of ear auricle (P <0.05) and foot swelling of rats (P <0.05, P <0.01). Significantly inhibited the expression of TNF-α, IFN-γ and IL-8 in serum of gouty arthritis rats (P <0.05, P <0.01), attenuated inflammatory cell infiltration, hyperemia, swelling and necrosis (P <0.05, P <0.01). Conclusion Jinling Tongfengshuwei Pill has a significant anti-inflammatory effect on acute inflammation. Its anti-inflammatory mechanism on gouty arthritis may be through the inhibition of the expression of pro-inflammatory cytokines TNF-α, IFN-γ and IL-8, Synovial tissue inflammatory response chain, which play an anti-inflammatory effect.
