为了研究骨中α粒子的微观分布,需进行骨切片制备。传统的脱钙骨切片,由于在脱钙过程中会丢失部分核素,从而不能如实地反映α粒子的微观分布和进行准确的计算.本文建立了不脱钙骨切片制备方法。详述了从(大)小白鼠股骨的获取、固定、脱水、浸透、包埋、到用西德产的Leitz 1600锯式切片机切片、染色一系列过程。通过摸索实验中各步骤的条件,制备出厚约30μm完整的不脱钙骨切片。
In order to study the microscopic distribution of alpha particles in bone, preparation of bone slices is required. Traditional decalcification bone slices will lose some of the nuclide during the decalcification process, which can not accurately reflect the microscopic distribution of α particles and perform accurate calculations. This paper established the method of preparation of non-decalcified bone slices. Details were obtained from the acquisition, fixation, dehydration, soaking, embedding of femurs from (large) mice and to a series of processes using a Leitz 1600 saw slicer produced in West Germany. By experimenting with the conditions of each step of the experiment, intact non-decalcified bone slices of about 30 μm in thickness were prepared.
