目的对壮药战骨种质资源的遗传多样性进行研究。方法采用ISSR分子标记技术分析9个战骨种群的遗传多样性。结果 13条引物共检测到73个位点,其中65个位点具有多态性,多态位点百分率(PPL)为89.04%。战骨总的Nei’s基因多样度指数(H)为0.226 5,Shannon’s多样性信息指数(I)为0.357 2,各种群的平均H、I为0.147 6、0.221 9。种群间基因分化系数(Gst)为0.348 4,基因流(Nm)为0.935 1。9个种群可聚类划分为2个大种群组。结论战骨种群遗传多样性为中等偏低水平;战骨种群的遗传变异主要存在于种群内;战骨的生物学特性和分布区域的片段化是导致种群间出现一定的遗传分化和种群遗传多样性较低的主要原因。
Objective To study the genetic diversity of Zhuangjiao Bone Germplasm Resources. Methods ISSR molecular markers were used to analyze the genetic diversity of nine bony populations. Results A total of 73 loci were detected by 13 primers, of which 65 loci were polymorphic and the percentage of polymorphic loci (PPL) was 89.04%. The total Nei’s gene diversity index (H) was 0.226 5 for bones and Shannon’s diversity index (I) was 0.357 2, and the average H of each group was 0.147 6,0.221 9. The coefficient of genetic differentiation (Gst) was 0.348 4 and the gene flow (Nm) was 0.935. The 1.9 populations were clustered into two large populations. CONCLUSIONS: The genetic diversity of the bony population is low to moderate level. The genetic variation of the bony population mainly exists in the population. The biological characteristics of the bones and the fragmentation of the distribution area lead to some genetic differentiation and population genetic diversity among the populations The main reason for low sex.
