探讨miR-581对结直肠癌SW620细胞侵袭转移能力的影响。首先,用miR-581mimics转染SW620细胞作为实验组(miR-581),用对照组mimics转染SW620细胞作为对照组(vector);用qRT-PCR检测2组细胞中miR-581的mRNA表达水平;划痕实验和transwell实验检测2组细胞迁移和侵袭能力的变化;Western blotting检测2组细胞中MMP-1和MMP-9的蛋白表达水平。结果显示,实验组细胞中miR-581表达较对照组明显增高(P<0.05);划痕实验显示实验组细胞划痕愈合率较对照组减慢(P<0.05),transwell结果显示实验组细胞较对照组细胞穿膜细胞数减少(P<0.05);Western blotting结果显示实验组MMP-1和MMP-9的表达都较对照组降低(P<0.05)。以上结果提示miR-581可以抑制SW620细胞的侵袭和转移。
To investigate the effect of miR-581 on invasion and metastasis of colorectal cancer SW620 cells. First, SW620 cells were transfected with miR-581mimics as experimental group (miR-581) and SW620 cells transfected with mimics as control; qRT-PCR was used to detect the mRNA expression level of miR-581 Scratch and transwell assays were used to detect the migration and invasion ability of the two groups. Western blotting was used to detect the protein expression of MMP-1 and MMP-9 in the two groups. The results showed that the expression of miR-581 in the experimental group was significantly higher than that in the control group (P <0.05). The scratch test showed that the wound healing rate of the experimental group was slower than that of the control group (P <0.05) Compared with the control group, the number of cell penetrating cells decreased (P <0.05). The results of Western blotting showed that the expression of MMP-1 and MMP-9 in the experimental group was lower than that in the control group (P <0.05). The above results suggest that miR-581 can inhibit the invasion and metastasis of SW620 cells.
