目的:建立同时测定乌药中4种生物碱的HPLC法。方法:Agilent ZORBAX SB-C18色谱柱(4.6 mm×250 mm,5μm);流动相A相为水(先加入1.5‰的二乙胺,再用冰醋酸调pH 3.0),B相为乙腈,线性梯度洗脱;UV检测波长289 nm。结果:波尔定碱在0.428~8.56μg,降波尔定碱在2.122~31.83μg,网状香荔枝碱在0.760~15.20μg,linderegatine在0.020 4~0.400 8μg与峰面积呈良好的线性关系,平均加样回收率为99.18%,100.94%,100.31%,99.17%,RSD分别为1.7%,2.5%,2.1%,3.0%。结论:该方法快速简便、准确,可用于乌药的质量评价。
Objective: To establish a HPLC method for simultaneous determination of four alkaloids in Radix Taraxaci. Methods: The Agilent ZORBAX SB-C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase A was water (first added 1.5 ‰ of diethylamine and then adjusted to pH 3.0 with glacial acetic acid) Gradient elution; UV detection wavelength 289 nm. Results: The results showed that there was a good linear relationship between the concentration of orally administered orally and the peak area of 0.428-8.56μg of orally administered ergine, 2.122-31.83μg of dilperidine, 0.760-15.15μg of reticuline and 0.020 4-0.400 8μg of linderegatine, The average recoveries were 99.18%, 100.94%, 100.31% and 99.17%, respectively, with RSDs of 1.7%, 2.5%, 2.1% and 3.0%, respectively.
