目的:通过分析5种内源激素GA3,IAA,ABA,ZR和JA在山药试管块茎形成建成中的动态变化,初步探讨山药试管块茎形成的生理生化机制。方法:山药试管苗在试管块茎诱导培养基MS+6-BA 1.5 mg.L-1+NAA 1.5 mg.L-1+蔗糖5%中培养,取不同生长期的小块茎,采用ELISA法对其内源激素进行分离、纯化、测定。结果:在山药试管块茎形成过程中,GA3含量呈现倒“S”形变化;IAA含量总体呈下降趋势;ABA,ZR初期含量升高,后期下降;JA含量则一直上升;GA3与ABA和JA比值变化较为显著。结论:在山药试管块茎形成过程中,内源激素ABA,JA,IAA,ZR和GA3起着重要的调控作用。
OBJECTIVE: To investigate the physiological and biochemical mechanisms of the tuber formation of yam tubers by analyzing the dynamic changes of five endogenous hormones, GA3, IAA, ABA, ZR and JA, in the formation of yam tubers. Methods: The yam test-tube plantlets were cultured in tuber-inducing medium MS + 6-BA 1.5 mg.L-1 + NAA 1.5 mg.L-1 + sucrose 5% in vitro. Small tubers with different growth stages were obtained. Endogenous hormones were isolated, purified and assayed. Results: In the process of tuber formation of Yam, the content of GA3 showed “S” shape change; the content of IAA decreased generally; the content of ABA and ZR increased at the early stage and decreased at the later stage; the content of GA3 increased with ABA and ABA JA ratio changes more significant. Conclusion: The endogenous hormones ABA, JA, IAA, ZR and GA3 play an important regulatory role in the tuber formation of yam.
