棉花黄萎病菌致萎峰蛋白氨基酸组分及其有关生化特性分析

利用快速蛋白质液相色谱技术 (简称 FPL C技术 )对 3个典型黄萎病菌系 T9、VD8和 SS-4的外泌毒蛋白进行层析分离 ,皆得到两个致萎蛋白峰 :峰 致萎力弱 ,棉苗受害症状属黄斑型 ;峰 致萎力极强 ,症状属青枯型。 3个菌系强致萎峰 毒蛋白的百分含量由高到低依次为 T9、VD8、SS-4。对各菌系致萎峰蛋白做氨基酸组分测定 ,结果显示 :13个菌系的致萎峰 毒蛋白中的碱性组氨酸百分含量很高 ,平均达 14 .76% ,远大于峰 毒蛋白中的组氨酸含量 ,其平均值仅 3 .74 % ;2 3个菌系各致萎峰皆测不出甲硫氨酸 ,酪氨酸仅在 T9的峰 中含 0 .13 % ,其余各峰含量均为零 ,各峰的半胱氨酸含量很低 ,都在 1%以下。还利用高效液相色谱技术估测致萎峰蛋白的分子量 ,并探讨了黄萎病菌致萎毒蛋白的有关结构功能特征。 Chromatographic separation of the exocrine proteins of three typical Verticillium dahliae strains T9, VD8 and SS-4 using fast protein liquid chromatography (FPL C) revealed two peaks of withering protein peaks Weak, the symptoms of cotton seedlings suffer from macular type; peak caused by extremely strong wilt symptoms are scape. The molecular weight of peaked protein was also estimated by HPLC, and the structural and functional characteristics of verticillin-wilt protein were also discussed.
