目的:探讨无能T细胞与凋亡的关系。方法:利用B71单抗和环孢素A(CsA)联用处理APC即T细胞系统,在体外诱致抗原特异性T细胞无能,用PI染色法经FACS分析了无能T细胞的凋亡,用RTPCR法检测了凋亡细胞FasmRNA的表达水平。结果:无能T细胞在1、3、5、10d细胞凋亡的百分率依次为2.16%、11.28%、19.27%、41.22%。当加入100U/mlIL2维持培养,无能T细胞未显示明显凋亡。无能T细胞活化后经PHA、PMA+A23187、CD3单抗再次诱导,24h便发生明显凋亡,凋亡百分率分别为34.64%、21.27%、15.07%,活化后的T细胞及无能T细胞凋亡时FasmRNA上调表达。结论:无能T细胞在体外是逐渐发生凋亡的,Fas/FasL参与细胞凋亡过程
Objective: To investigate the relationship between incompetent T cells and apoptosis. Methods: The combination of B71 monoclonal antibody and cyclosporine A (CsA) treatment of APC or T cell system in vitro induced antigen-specific T cell incompetence, PI staining by FACS analysis of incompetent T cell apoptosis, The expression of Fas mRNA in apoptotic cells was detected by RT-PCR. Results: The percentages of apoptosis of incompetent T cells at 1, 3, 5 and 10 days were 2.16%, 11.28%, 19.27% and 41.22%, respectively. When adding 100U / ml IL 2 maintenance culture, incapable T cells showed no significant apoptosis. After activation of incompetent T cells, the cells were induced again by PHA, PMA + A23187 and CD3 monoclonal antibody, and obvious apoptosis occurred at 24h, with the percentages of apoptosis being 34.64%, 21.27% and 15.07%, respectively. The activated T cells and impotence T-cell apoptosis FasmRNA up-regulated expression. CONCLUSION: Incomplete T cells are gradually apoptotic in vitro, and Fas / FasL is involved in the process of apoptosis
