麦胚提取物对成骨细胞TGF-β1和IGF-1表达的影响

目的 通过研究成骨细胞中转化生长因子 (TGF) - β1 和胰岛素样生长因子(IGF) - 1的表达 ,进一步探讨麦胚乙醇提取物对大鼠成骨细胞的作用机理。方法 体外培养新生大鼠颅骨成骨细胞 ,用免疫组化方法测定成骨细胞中TGF β1 和IGF 1的表达情况 ,用HPIAS - 1 0 0 0图象分析仪对各组表达进行定量分析。结果 麦胚提取物培养的大鼠第 3代成骨细胞的TGF β1 和IGF 1呈阳性表达 ,麦胚提取物组的TGF - β1 和IGF 1平均光密度都显著高于阴性对照组 (P <0 .0 5 ,P <0 .0 1 ) ,但与阳性对照组比较无显著差异 (P >0 .0 5)。结论 麦胚乙醇提取物可增强新生大鼠成骨细胞TGF β1 和IGF 1的表达 OBJECTIVE: To investigate the mechanism of ethanol extract of rat embryos on rat osteoblasts by studying the expression of transforming growth factor (TGF) - β1 and insulin - like growth factor (IGF - 1) in osteoblasts. Methods The osteoblasts of neonatal rat calvaria were cultured in vitro. The expression of TGF β1 and IGF 1 in osteoblasts were detected by immunohistochemistry. The expression of TGF β1 and IGF 1 in osteoblasts was analyzed by using HPIAS - 1 000 image analyzer. Results The expression of TGFβ1 and IGF - 1 in 3rd generation of osteoblasts cultured in wheat germ extract was positive. The average optical density of TGF - β1 and IGF - 1 in wheat germ extract group was significantly higher than that in negative control group (P < 0. 05, P <0. 01), but no significant difference compared with the positive control group (P> 0.05). Conclusion Wheat germ ethanol extract can enhance the expression of TGF β1 and IGF 1 in osteoblasts of neonatal rats
