目的探讨调节性T细胞(regulatory T cell,Treg)在年轻女性宫颈鳞状上皮内病变(cervical squamous intraepithelial lesions,CSIL)、宫颈癌组织中的表达及其与不同病变级别的相关性,并研究其预测CSIL转归的作用。方法选取2014年6月-2016年6月在广东省妇幼保健院或深圳市妇幼保健院就诊的年龄<35岁CSIL、宫颈癌患者110例,分为宫颈低级别鳞状上皮内病变(Low-grade squamous intraepithelial lesion,LSIL)组(30例)、宫颈高级别鳞状上皮内病变(High-grade squamous intraepithelial lesion,HSIL)组(30例)及宫颈癌组(50例),另选取同期因其他疾病(如子宫肌瘤、子宫内膜异位症)行全子宫切除的正常宫颈组织30例为正常对照组。进行免疫组织化学染色,检测宫颈组织中叉头状旋转录因子3(forkhead helix transcription,FOXP3)的表达情况。结果 FOXP3在正常对照组、LSIL组、HSIL组及宫颈癌组中的阳性表达率分别为3.3%、36.6%、53.3%和100.0%,FOXP3在不同级别CSIL、宫颈癌中的总阳性率有显著差别,疾病级别越重,总阳性率越高。正常对照组、LSIL组、HSIL组及宫颈癌组病变组织中,FOXP3的阳性表达强度与病变级别相关(P<0.001)。结论 FOXP3在年轻女性CSIL、宫颈癌组织中阳性表达强度与病变级别相关,可辅助临床评价肿瘤的生物学行为。
Objective To investigate the expression of regulatory T cell (Treg) in cervical squamous intraepithelial lesions (CSIL) and cervical cancer and its relationship with different pathological grades, Predict the outcome of CSIL. Methods From June 2014 to June 2016, 110 patients with CSIL and cervical cancer who were treated at Guangdong Maternal and Child Health Hospital or Shenzhen Maternal and Child Health Hospital were selected and divided into low-grade cervical low- grade squamous intraepithelial lesion (LSIL) group (30 cases), high-grade squamous intraepithelial lesion (HSIL group) (30 cases) and cervical cancer group (50 cases) Diseases (such as uterine fibroids, endometriosis) underwent a total hysterectomy of normal cervical tissue 30 cases of normal control group. Immunohistochemical staining was performed to detect the expression of forkhead helix transcription (FOXP3) in cervical tissue. Results The positive rates of FOXP3 in normal control group, LSIL group, HSIL group and cervical cancer group were 3.3%, 36.6%, 53.3% and 100.0%, respectively. The positive rates of FOXP3 in different grades of CSIL and cervical cancer were significant Differences, the more severe the disease, the higher the total positive rate. In normal control group, LSIL group, HSIL group and cervical cancer group, the positive expression of FOXP3 was correlated with the grade of lesions (P <0.001). Conclusions The positive expression level of FOXP3 in young female CSIL and cervical cancer is related to the grade of the lesion. It can help clinical evaluate the biological behavior of the tumor.
