本文研究了60Gy照射的胸腺细胞滋养层对小鼠骨髓细胞体外单层培养CFU-F成集落的影响,胸腺滋养层细胞对低密度骨髓细胞(24孔培养板每孔3×10~4个)无成集落刺激作用,而对高密度骨髓细胞(2×10~5个/孔)则有成集落促进作用。添加滋养层细胞密度在10~5~10~6个/cm~2条件下,随着滋养细胞的增多,小鼠骨髓CFU-F成集落数也增加,甚至可达到集落融合生长的程度。本结果为进一步研究造血基质祖细胞、造血免疫器官损伤修复及免疫对造血影响创造了好的方法。
In this paper, the effects of 60 Gy irradiation of thymus trophoblast cells on the monolayer CFU-F colonies of mouse bone marrow cells in vitro were studied. Thymus trophoblast cells were cultured on low density bone marrow cells (3 × 10 ~ 4 per 24 well plate) No colony stimulating effect, while the high-density bone marrow cells (2 × 10 ~ 5 / hole) is a colony-promoting effect. Under the condition of adding trophoblast cell density of 10 ~ 5 ~ 10 ~ 6 / cm ~ 2, with the increase of trophoblast cells, the number of CFU-F into the bone marrow also increased, and even reached the level of colony fusion growth. The results provide a good method for further study of hematopoietic progenitor cells, hematopoietic organ damage repair and immune effects on hematopoiesis.
