目的:建立人肾细胞癌原位移植裸鼠肺转移动物模型,研究Has mir-129对肾脏移植瘤和肺部转移瘤的影响。方法:构建Has-mir-129慢病毒载体,以人肾癌细胞株SN12-PM6建立的动物模型作为对照组,以Has-mir-129慢病毒载体感染肾癌细胞建立的动物模型作为实验组;7周后观察裸鼠肾脏肿瘤及肺部转移瘤的生长情况,并分别计算两组的肾脏成瘤率、肺部转移率及肿瘤的重量;所有标本均采用10%甲醛溶液固定,常规石蜡包埋、切片、HE染色后分析结果。结果:对照组肾脏成瘤率为100%(8/8),肺部转移率为100%(8/8),肿瘤重量平均为(0.712 8±0.147 4)g;实验组肾脏成瘤率为100%(8/8),肺部转移率为62.5%(5/8),肿瘤重量平均为(0.481 5±0.111 2)g;肾脏成瘤率两者之间差异无统计学意义,肿瘤重量和肺部转移率差异有统计学意义(P<0.05)。结论:成功建立了Has-mir-129慢病毒载体及人肾细胞癌原位移植裸鼠肺转移动物模型,发现Has-mir-129能抑制人肾细胞癌原位移植裸鼠肺转移模型移植瘤的生长及肺部转移瘤的形成。
OBJECTIVE: To establish animal model of orthotopic lung transplantation in human renal cell carcinoma in nude mice and to study the effect of Has mir-129 on renal xenografts and lung metastases. Methods: The Has-mir-129 lentiviral vector was constructed. The animal model of human renal cell carcinoma cell line SN12-PM6 was used as the control group. The animal model established by the infection of Has-mir-129 lentivirus vector with renal cancer cells was used as the experimental group. After 7 weeks, the growth of renal tumors and metastatic tumors of the lung in nude mice were observed. The rates of neoplasia, lung metastasis and tumor weight were calculated in both groups. All samples were fixed with 10% formaldehyde solution and paraffin Buried, sliced, HE staining analysis of the results. Results: The tumor formation rate of kidney in control group was 100% (8/8), the lung metastasis rate was 100% (8/8), the average tumor weight was (0.712 8 ± 0.147 4) g. 100% (8/8), lung metastasis rate was 62.5% (5/8), the average tumor weight was (0.481 5 ± 0.111 2) g; There was no significant difference between the two groups in tumor formation rate And pulmonary metastasis rate difference was statistically significant (P <0.05). Conclusion: Has-mir-129 lentiviral vector and human renal cell carcinoma orthotopically transplanted animal model of lung metastasis in nude mice were successfully established.
