目的:探讨肿瘤坏死因子(TNF)和化疗药物联合应用的作用机制。方法:用3AO卵巢癌细胞株作为靶细胞,体外培养,分单独用药及联合用药组,于用药后不同时间收获细胞,制成电镜标本,在透视电镜下观察单独应用TNF及TNF与化疗药物(阿霉素、更生霉素)联合用药对卵巢癌细胞超微结构的改变。结果:电镜显示,单独应用TNF,卵巢癌细胞无明显改变,联合用药后靶细胞改变出现时间早,程度重,主要在细胞质,表现为线粒体水肿、嵴消失,内质网增生、扩张,细胞空泡样变性多见,而细胞膜变化不明显。结论:TNF与化疗药物有协同抗卵巢癌细胞的作用
Objective: To explore the mechanism of the combined application of tumor necrosis factor (TNF) and chemotherapeutics. METHODS: 3AO ovarian cancer cell lines were used as target cells, cultured in vitro, divided into single drug and combination groups. Cells were harvested at different times after the administration to prepare electron microscopy specimens. TNF and TNF alone and chemotherapeutic drugs were observed under transmission electron microscope ( The combination of adriamycin and dactinomycin on the ultrastructural changes of ovarian cancer cells. RESULTS: The results of electron microscopy showed that ovarian cancer cells had no significant changes when TNF alone was used. Changes in target cells after the combination treatment were early and severe, mainly in the cytoplasm, showing mitochondrial edema, loss of iliac crest, proliferation, expansion of endoplasmic reticulum, and cell emptying. Follicular degeneration was more common, but cell membrane changes were not obvious. Conclusion: TNF and chemotherapeutics have synergistic effect on ovarian cancer cells
