目的:探讨氟伐他汀(fluvastatin,Flu)对腹膜透析(peritoneal dialysis,PD)大鼠腹膜组织形态影响的初步研究。方法:通过SD雄性大鼠(体重180~200 g)腹腔注射4.25%浓度的葡萄糖腹透液的方式模拟腹膜透析。随机分为5个组:①正常对照组;②4.25%腹透液组;③单纯Flu组;④生理盐水组;⑤Flu干预的4.25%腹透液组。4周后留取各组大鼠壁层腹膜组织,光镜观察各组大鼠壁层腹膜形态学的变化,同时采用RT-PCR法测定各组大鼠腹膜转化生长因子-β1(transforming growth factor,TGF-β1)及纤维连接蛋白(fibronectin,FN)mRNA的表达。结果:光镜下HE染色显示4.25%腹透液组大鼠腹膜较正常对照组显著增厚,Flu干预组的壁层腹膜厚度薄于未干预组,生理盐水组和单纯Flu组大鼠腹膜的形态较正常对照组未见明显改变。4.25%腹透液组大鼠腹膜TGF-β1及FN mRNA的表达量均高于正常对照组(均P<0.05),Flu干预组两者mRNA的表达量均明显低于未干预组(均P<0.05),生理盐水组和单纯Flu组大鼠腹膜两者的mRNA表达量较正常组无明显统计学意义。结论:长期使用高糖腹透液会改变腹膜的结构,氟伐他汀在一定程度上对腹膜有保护作用,但具体机制仍需进一步研究。
Objective: To investigate the effect of fluvastatin (Flu) on peritoneal histomorphology of peritoneal dialysis (PD) rats. METHODS: Peritoneal dialysis was simulated by intraperitoneal injection of 4.25% glucose dialysate into SD male rats weighing 180-200 g. Randomly divided into 5 groups: ① normal control group; ② 4.25% dialysate group; ③ Flu group alone; ④ saline group; Four weeks later, the peritoneum of the parietal peritoneum in each group was collected. The peritoneal morphological changes in the parietal layer were observed with light microscope. The expressions of transforming growth factor-β1 (TGF-β1) , TGF-β1) and fibronectin (FN) mRNA expression. Results: HE staining showed that the peritoneum of 4.25% PD group was significantly thicker than that of the normal control group, and the thickness of the parietal peritoneum of Flu intervention group was thinner than that of the non-intervention group. In the normal saline group and the pure Flu group, Morphology than the normal control group no significant change. The expression of TGF-β1 and FN mRNA in peritoneum of 4.25% PD group was significantly higher than that of normal control group (all P <0.05), and the expression of both mRNA in Flu intervention group was significantly lower than that in non-intervention group (P <0.05). The mRNA expression of both peritoneum in normal saline group and pure Flu group had no statistical significance compared with normal group. Conclusion: Long-term use of high-sugar peritoneal dialysis will change the structure of the peritoneum. Fluvastatin may have a protective effect on the peritoneum to a certain extent, but the specific mechanism still needs further study.
