棉花生长后期进行种间杂交和杂交胚珠离体培养初报

以陆地棉华棉4号为母本,与随县中棉进行杂交,授粉后48小时开始取材培养,同时设置三个对照。杂交留株生长的全部随钤脱落,经离体培养的却获得11%的成胚率。不同培养基上离体培养幼胚的效果很不相同。适量的激素配合对杂交胚珠的培养表现良好效果,赤毒素对珠被和纤维的发育有明显的促进作用,但纤维生长与否并不能作为胚是否发育的依据。随着培养间时的延长,培养基PH值逐渐向碱性变化,这一变化对幼胚发育有明显的阻碍作用。本试验选择在棉花生长后期进行,以期延长取材和培养工作时间以及在更大范围内提高培养效果。 The cotton Huameian 4 was used as the female parent and crossed with the cotton of Sui County. The materials were harvested 48 hours after pollination, and three controls were set at the same time. All strains of hybrid plants grow with the shedding, but the obtained in vitro culture rate of 11% embryos. The effects of in vitro culture of immature embryos on different media are very different. The appropriate amount of hormone cooperates well to the culture of hybrid ovules, and the toxin has obvious effect on the development of the beads and fiber, but the fiber growth can not be used as the basis for the embryo development. With the extension of culture time, the pH value of the medium gradually changed to alkaline, and this change had a significant hindrance to the development of immature embryos. The experiment was conducted in the later stage of cotton growth in order to prolong the working time of material collection and cultivation and to improve the culture effect in a wider range.
