目的:通过构建2型糖尿病(T2DM)大鼠妊娠模型,探讨左归复方对2型糖尿病妊娠大鼠模型胰腺组织CD4、TGF-β1表达的影响。方法:将40只雌性SD大鼠随机分为5组,其中4组以高糖高脂饮食并腹腔注射链脲佐菌素(STZ)建立2型糖尿病大鼠模型组,采用电化学法对各组大鼠按照血糖水平排序,随机将40只雌性SD大鼠分为正常组、中药低剂量组、中药高剂量组、达美康组、模型组,2∶1雌雄比例合笼,建立2型糖尿病大鼠妊娠模型。观察大鼠一般情况,妊娠第0.5天开始对以上5组分别予等容量0.9%氯化钠注射液、低剂量左归复方、高剂量左归复方、达美康混悬液、蒸馏水灌胃21d,观察各组大鼠胰腺组织病理形态学的变化,免疫组化法检测胰腺CD4、TGF-β1蛋白的表达水平。结果:与正常对照组比较,模型组胰腺CD4水平显著升高,TGF-β1水平显著降低,差异均有统计学意义(P<0.01或P<0.05);与模型组比较,中药低、高剂量组胰腺CD4水平显著降低,达美康组TGFβ1水平显著升高,差异有统计学意义(P<0.01或P<0.05);中药低剂量组较达美康组CD4、TGFβ1水平低,差异有统计学意义(P<0.01)。结论:左归复方能够改善2型糖尿病大鼠CD4、TGF-β1蛋白的表达。
Objective: To investigate the effects of Zuogui Recipe on the expression of CD4 and TGF-β1 in the rat model of type 2 diabetes mellitus by establishing pregnancy model of type 2 diabetes mellitus (T2DM) rats. Methods: Forty female Sprague-Dawley rats were randomly divided into five groups. Four groups were given type 2 diabetes mellitus with high glucose and high fat diet and intraperitoneal injection of streptozotocin (STZ) The rats were randomly divided into normal group, low dose Chinese medicine group, high dose Chinese medicine group, daimikang group, model group and male and female ratio of 2: 1. Diabetic rat pregnancy model. Observed the general situation in rats, the first 0.5 days of pregnancy to the above five groups were given the same volume of 0.9% sodium chloride injection, low-dose left Zuoguifu, high-dose Zuoguifu side, up to the United States Kang suspension, The morphological changes of pancreas in each group were observed. The expression of CD4, TGF-β1 protein in pancreas was detected by immunohistochemistry. Results: Compared with the normal control group, the levels of CD4 in the pancreas and the levels of TGF-β1 in the model group were significantly decreased (P <0.01 or P <0.05). Compared with the model group, The level of CD4 in the pancreas decreased significantly, and the level of TGFβ1 in the control group was significantly increased (P <0.01 or P <0.05). The levels of CD4 and TGFβ1 in the low dose group were lower than those in the control group Significance (P <0.01). Conclusion: Zuogui Recipe can improve the expression of CD4 and TGF-β1 in type 2 diabetic rats.
