目的建立HPLC测定铁棒锤中乌头碱的方法。方法采用高效液相色谱-紫外检测器;色谱柱为Thermo ODS-2 HYPERSIL Dim(.250mm×4.6mm,5μm);柱温为30℃;流动相为乙腈-0.3%二乙胺水溶液(35:65);流速为1.0ml/min。结果乌头碱在0.975μg-3.9μg(R=0.999)范围内线性关系良好。乌头碱加样回收率为98.06%,RSD%为1.2%,原药材中乌头碱的含量为0.1052mg/g。结论本实验建立的测定方法具有准确、简便、灵敏度高、无干扰而且重现性好的优点,适合用于铁棒锤中乌头碱含量的测定。
Objective To establish a method for the determination of aconitine in iron hammer hammer by HPLC. Method: HPLC-UV detector; Thermo ODS-2 HYPERSIL Dim (.250mm×4.6mm, 5μm) column; column temperature 30°C; mobile phase acetonitrile-0.3% diethylamine solution (35: 65); flow rate 1.0 ml/min. Results The aconitine had good linearity in the range of 0.975μg-3.9μg (R=0.999). The recovery rate of aconitine was 98.06%, RSD% was 1.2%, and the content of aconitine in the original drug was 0.1052 mg/g. Conclusion The assay established in this experiment has the advantages of accuracy, simplicity, high sensitivity, no interference and good reproducibility, and is suitable for the determination of aconitine in iron hammer hammers.
