目的:研究低温冻存(cryopreservation)对牙髓干细胞(dental pulp stem cells,DPSCs)生物学特性的影响。方法:取牙髓组织进行干细胞分离培养,用流式细胞仪、细胞增殖检测以及多向诱导分化对比研究未低温冻存与冻存1年后复苏的DPSCs自我更新和多向分化潜能之间的差异。结果:低温冻存对DPSCs的干细胞表面标记分子、增殖以及成骨和成脂分化均无显著影响。结论:低温冻存对DPSCs生物学特性无显著影响,是长期进行干细胞储存的可行方法。
Objective: To study the effects of cryopreservation on the biological characteristics of dental pulp stem cells (DPSCs). Methods: The dental pulp tissue was isolated and cultured. The relationship between self-renewal and multidrug differentiation potential of non-cryopreserved and resuscitated DPSCs after cryopreservation was compared by flow cytometry, cell proliferation assay and multidirectional induced differentiation difference. RESULTS: Cryopreservation had no significant effect on the surface marker molecules, proliferation, osteogenic and adipogenic differentiation of DPSCs. Conclusion: Cryopreservation has no significant effect on the biological characteristics of DPSCs, which is a feasible method for long-term storage of stem cells.
