目的 比较糖尿病视网膜病变 (diabetic vitreoretinopathy,DR)及非糖尿病视网膜病变 (non-DR)患者玻璃体中基质金属蛋白酶 (m atrix metalloproteinase,MMP)表达及活性 ;并探讨 MMP表达的相关因素。 方法 以 31例 DR、17例 non- DR(9例黄斑裂孔 ,8例视网膜前膜 )玻璃体为样本 ,采用明胶酶谱(Zymography)技术检测样本中 MMP- 2、9,并用 Western印迹杂交方法证实 MMP- 2、9表达。 结果 所有样本中都检测到 MMP- 2表达 ,其中 DR玻璃体中 MMP- 2活性略高于 non- DR,但差别无显著性意义。45 .2 %的 DR玻璃体样本中有 MMP- 9表达 ,而 non- DR玻璃体样本中无一例有 MMP- 9表达。 DR玻璃体样本中 ,部分性玻璃体后脱离 (posterior vitreous detachment,PVD)者 MMP- 9表达的阳性率 (6 6 .7% )明显高于完全性 PVD者 (15 .4% ) (P<0 .0 1)。Western印迹杂交结果进一步证实了 MMP- 2、9表达。 结论 DR及 non- DR玻璃体样本中 MMP- 2活性无明显差异 ;non- DR玻璃体样本中未见 MMP- 9表达 ,而 DR玻璃体样本中 MMP- 9表达率与部分性 PVD密切相关。 MMP- 9可能与 DR的发生有关。
Objective To compare the expression and activity of matrix metalloproteinase (MMP) in vitreous with diabetic vitreoretinopathy (DR) and non-diabetic retinopathy (non-DR) and explore the related factors of MMP expression. Methods The vitreous of 31 cases of DR and 17 cases of non-DR (9 cases of macular hole, 8 cases of pre-retinal membrane) were taken as samples. Gelatin zymography was used to detect MMP-2 and 9 in samples. Western blotting was used to confirm the expression of MMP- MMP-2, 9 expression. Results The expression of MMP-2 was detected in all the samples. The activity of MMP-2 in DR vitreous was slightly higher than that in non-DR, but the difference was not significant. 45.2% of the DR vitreous samples had MMP-9 expression, while none of the non-DR vitreous samples had MMP-9 expression. In the DR vitreoretinal samples, the positive rate of MMP-9 expression in patients with partial vitreous detachment (PVD) was significantly higher than that in patients with complete PVD (15.4%) (P <0. 0 1). Western blot hybridization results further confirmed that MMP-2,9 expression. Conclusion There was no significant difference in the activity of MMP-2 between DR and non-DR vitreous samples. No expression of MMP-9 was found in non-DR vitreous samples. The expression of MMP-9 in DR vitreous samples was closely related to partial PVD. MMP-9 may be related to the occurrence of DR.
