目的:测定市售川乌不同品种中总黄酮的含量并比较其差异。方法:利用超声波提取法,在温度55℃、时间40min、以70%乙醇为提取溶剂、物料比为1∶20(m/V)的条件下提取10个样品中的黄酮类化合物;以芦丁为对照品,在510nm波长处,采用紫外分光光度法测定总黄酮的含量。结果:芦丁的质量浓度在0~0.0988mg/ml范围内与其吸光度呈良好的线性关系(r=0.9998);精密度、稳定性、重复性试验的RSD均<2%;平均加样回收率为97.93%,RSD=1.55%(n=6)。10个市售川乌样品中,总黄酮质量分数最高的是8号,其质量分数极显著高于10、4、6、7、2、1、9、3号,显著高于5号;3号川乌的总黄酮质量分数最低,但与7、2、1、9号川乌比较差异无统计学意义。结论:市售不同品种川乌中总黄酮的质量分数存在明显差异,临床应注意甄别使用。
Objective: To determine the content of total flavonoids in different varieties of Radix Aconiti commercially available and compare the differences. Methods: Flavonoids were extracted from 10 samples by ultrasonic extraction at 70 ℃ for 40 min and 70% ethanol as extraction solvent at a feed ratio of 1:20 (m / V) As a reference substance, at 510nm wavelength, the determination of total flavonoids by UV spectrophotometry. Results: The concentration of rutin showed a good linear relationship with the absorbance (r = 0.9998) in the range of 0 ~ 0.0988mg / ml. The RSD of precision, stability and repeatability were all less than 2%. The average recoveries 97.93%, RSD = 1.55% (n = 6). Among the 10 samples of Radix Chuanxiong, the highest content of total flavonoids was No. 8, the mass fraction of which was significantly higher than that of No. 10,4,6,7,2,1,9,3, which was significantly higher than that of No. 5; 3 Chuanwu the total flavonoids mass fraction lowest, but with 7,2,1,9 Chuanwu no significant difference. Conclusion: There is a significant difference in the mass fraction of total flavonoids from different varieties of Radix Aconiti commercially available. Clinical application should be screened.
