目的探讨不同吸烟周龄小鼠的精子活率和睾丸组织外周致密纤维1(ODF1)mRNA的表达。方法 6~8周龄的C57BL/6J健康雄性小鼠分别给予吸烟2周、4周和6周处理,ELISA试剂盒检测血清可替宁含量,血球计数板计数精子活率,实时荧光定量PCR检测ODF1mRNA表达水平的改变。结果经过2周、4周和6周的烟雾处理后,小鼠血清中可替宁含量明显高于不吸烟组,差异均有统计学意义(P<0.05)。吸烟6周后,小鼠血清中可替宁含量可达(10.76±2.19)ng/ml。此外,吸烟6周小鼠的睾丸组织ODF1mRNA表达量明显低于对照组(P<0.05)。经过烟雾处理后,小鼠精子活率并没有发生显著改变。结论本实验成功建立吸烟小鼠模型,可以为其他疾病的研究提供吸烟小鼠模型。其次,吸烟可能通过影响ODF1基因的表达而作用于精子的发生阶段,从而影响男性生殖系统。
Objective To investigate the sperm motility and the expression of ODF1 mRNA in testis of different smoking age mice. Methods C57BL / 6J healthy male mice aged 6 to 8 weeks were given smoking for 2 weeks, 4 weeks and 6 weeks respectively. Serum cotinine content, hemocytometer sperm count, and real-time fluorescence quantitative PCR ODF1 mRNA expression level changes. Results After two weeks, four weeks and six weeks of smoke treatment, the content of cotinine in mice serum was significantly higher than that in non-smoking group, the difference was statistically significant (P <0.05). Six weeks after smoking, the cotinine content in the mouse serum was up to (10.76 ± 2.19) ng / ml. In addition, the expression of ODF1 mRNA in testis tissue of 6-week smoking mice was significantly lower than that of the control group (P <0.05). After smoked, the mouse sperm motility did not change significantly. Conclusion This experiment successfully established smoking mouse model, which can provide smoking mice model for other diseases. Second, smoking may affect the male reproductive system by influencing the expression of ODF1 gene during the stage of spermatogenesis.
