目的:观察P-Akt和p27Kip1蛋白在鼻咽癌组织中的表达并分析两者之间的相互关系。方法:应用免疫组织化学技术(SABC)检测60例手术活检切取的鼻咽癌组织蜡块,与30例非肿瘤鼻咽组织石蜡标本作比较,分析鼻咽癌组织中磷酸化Akt(P-Akt)和p27Kip1蛋白的表达之间的相关性。结果:P-Akt蛋白在鼻咽癌中胞质表达高于非肿瘤鼻咽组织,P<0.001,p27Kip1蛋白在鼻咽癌细胞核表达较非肿瘤鼻咽组织降低,P<0.01;鼻咽癌细胞中P-Akt的表达与胞核p27Kip1的表达呈负相关,与胞质p27Kip1的表达呈正相关。结论:P-Akt蛋白过表达的鼻咽癌患者p27Kip1蛋白胞核表达降低,提示P-Akt蛋白和p27Kip1蛋白可能分别作为正性和负性调控因子,共同作用于细胞恶性增殖的过程。
Objective: To observe the expression of P-Akt and p27Kip1 in nasopharyngeal carcinoma and to analyze the relationship between the two. Methods: The immunohistochemistry (SABC) method was used to detect the paraffin blocks of 60 cases of nasopharyngeal carcinoma excised from surgical biopsies and compared with 30 cases of non-tumor nasopharyngeal paraffin specimens. The expressions of phosphorylated Akt (P-Akt ) And p27Kip1 protein expression. Results: The expression of P-Akt protein in nasopharyngeal carcinoma was higher than that in non-tumor nasopharyngeal tissue (P <0.001). The expression of p27Kip1 in nasopharyngeal carcinoma cells was lower than that in non-tumor nasopharyngeal tissues (P <0.01) The expression of P-Akt was negatively correlated with the expression of p27Kip1 in the nucleus and positively correlated with the expression of p27Kip1 in the cytoplasm. Conclusion: The decreased expression of p27Kip1 in NPC cells with P-Akt protein overexpression suggests that P-Akt protein and p27Kip1 protein may serve as positive and negative regulators, respectively, and play a role in the malignant proliferation of cells.
