目的建立LC-MS/MS法测定红景天苷在大鼠血浆中的浓度。方法色谱柱:Agela C18柱(4.6 mm×150 mm,5μm),流动相:乙腈-水(15∶85),流速:1 mL·min-1,用沉淀蛋白法进行血浆样品预处理,以多反应监测(MRM)扫描方式检测,表告依春为内标。结果血浆中红景天苷质量浓度在0.04~80.00μg·mL-1内线性关系良好(r=0.9947),最低定量限为0.04μg·mL-1,日内精密度RSD≤4.5%,日间精密度RSD≤10.6%,平均提取回收率为(90.1±4.6)%(n=3),基质效应为(99.3±7.8)%(n=3)。结论建立的LC-MS/MS法简便、快捷、准确,能满足红景天苷的血药浓度测定及其药代动力学研究。
Objective To establish a LC-MS / MS method for the determination of salidroside in rat plasma. Methods The chromatographic column was composed of Agela C18 column (4.6 mm × 150 mm, 5 μm), mobile phase of acetonitrile-water (15:85), flow rate of 1 mL · min-1 and pretreated plasma samples Response monitoring (MRM) scan detection, the report according to spring as the internal standard. Results The plasma concentration of salidroside in 0.04 ~ 80.00μg · mL-1 within a good linear relationship (r = 0.9947), the lowest limit of quantification of 0.04μg · mL-1, the intra-day precision RSD ≤ 4.5% day precision RSD≤10.6%, the average recovery was (90.1 ± 4.6)% (n = 3) and the matrix effect was (99.3 ± 7.8)% (n = 3). Conclusion The established LC-MS / MS method is simple, rapid and accurate and can meet the requirements of salidroside in plasma concentration determination and pharmacokinetics.
