为了探究黄连水煎液对小鼠脑组织急性缺血再灌注损伤的保护作用,我们将50只小鼠随机分为5组分别是:假手术对照组、模型对照组(用玻璃分针分离两侧颈总动脉和迷走神经,结扎法同时结扎颈总动脉,造成小鼠急性脑组织缺血再灌注损伤)、黄连水煎液治疗组(25 mg/m L,50 mg/m L,100 mg/m L)。各治疗组分别灌胃给予上述浓度黄连水煎液,模型对照组和假手术对照组给予生理盐水,上述各组均灌胃7 d;在灌胃结束后,断头取出脑组织,制备脑组织匀浆,测定脑组织中Evan’s蓝含量及抗氧化酶(SOD)活性与丙二醛(MDA)含量的变化。结果表明:缺血再灌注后小鼠脑组织Evan’s蓝含量升高,SOD活性降低,MDA含量增高。用药后小鼠脑组织Evan’s蓝含量降低,SOD活性升高,MDA含量降低。因此,黄连水煎液能改善小鼠脑组织缺血区的血流量,减轻急性脑缺血的损伤,对脑缺血再灌注损伤有保护作用。

The results showed that the content of Evan’s blue, the activity of SOD and the content of MDA increased in the brain of mice after ischemia-reperfusion. Evan’s blue content decreased, SOD activity increased and MDA content decreased in mice brain after treatment. Therefore, Coptis decoction can improve cerebral blood flow in mice ischemic area, reduce acute cerebral ischemia injury, cerebral ischemia-reperfusion injury has a protective effect.
