目的探讨槲皮素对宫颈癌侵袭和转移的作用机制及其与乙酰肝素酶(heparanase,HPA)的相关性。方法构建宫颈癌Hela细胞组、Hela+槲皮素组、HPA-SiRNA-Hela组、HPA-SiRNA-Hela+槲皮素组4组细胞模型,应用Transwell小室基质划痕损伤实验方法,测定不同时间点各组宫颈癌Hela细胞侵袭能力的变化。结果在6,12,24h时,Hela+槲皮素组及HPA-SiRNA-Hela组与Hela细胞组比较,宫颈癌Hela细胞迁移能力明显下降(P<0.01);HPA-SiRNA-Hela组与Hela+槲皮素组比较差异无统计学意义(P>0.05);HPA-SiRNA-Hela+槲皮素组与其他3组比较,宫颈癌Hela细胞迁移能力均下降(P均<0.01)。结论槲皮素对宫颈癌Hela细胞的侵袭能力有明显抑制作用,HPA与宫颈癌Hela细胞的侵袭过程相关,而槲皮素对宫颈癌Hela细胞的侵袭能力的抑制作用可能与HPA相关。
Objective To investigate the mechanism of quercetin on invasion and metastasis of cervical carcinoma and its relationship with heparanase (HPA). Methods Hela cells, Hela + quercetin, HPA-SiRNA-Hela and HPA-SiRNA-Hela + quercetin groups were used in this study. Transwell assay of cell matrix scratch was used to determine the cell cycle arrest Changes of invasiveness of cervical cancer Hela cells. Results The migration of Hela cells was significantly decreased in Hela + quercetin and HPA-SiRNA-Hela groups compared with Hela cells at 6, 12 and 24 hours (P <0.01). The HPA-SiRNA-Hela and Hela + (P> 0.05). Compared with the other three groups, the migration ability of Hela cells in HPA-SiRNA-Hela + quercetin group was decreased (all P <0.01). Conclusions Quercetin can significantly inhibit the invasion of cervical cancer Hela cells. HPA is associated with the invasion of cervical cancer Hela cells. The inhibitory effect of quercetin on the invasion of cervical cancer Hela cells may be related to HPA.
