最近,美国的Genentech与Genetics Institute的二组科学家同时成功地克隆了凝血因子Ⅷ∶C的DNA,并使其在哺乳类细胞株中表达产生有活性的因子Ⅷ∶C。他们根据已纯化的来源于猪或人因子Ⅷ∶C的一小片段氨基酸序列,合成出寡核苷酸探针,用此探针从基因库中找出了因子Ⅷ的基因,并已弄清楚这段基因总长为186000bp,共有长度为69~3106bp不等的26个外显子,整个基因约占X染色体的0.1%。他们分别利用人的T细胞杂交瘤株或人肝获得了cDNA克隆,并根据cDNA的序列分析推断出成熟蛋白的2332个氨基酸序列。结果显示,多肽链中有与凝血因子V结构同源的区域以及与血浆铜兰蛋白结
