目的研究犬颅脑枪弹伤后脑组织中脑源性神经生长因子(BDNF)和怏反应蛋白c-myc的变化规律。方法24只杂种犬随机分为对照组(n=4),枪弹伤组(n=20)。采用德国小口径步枪子弹致犬颅脑额叶切线伤。用免疫组织化学方法检测伤后0.5、2、6、12和24 h各不同时期弹道挫伤区、震荡区、海马及脑干神经元中BDNF和c-myc蛋白的表达。结果对照组脑神经元中BDNF和c-myc蛋白表达弱。枪弹伤组挫伤区、震荡区、海马及脑干神经元中BDNF和c-myc蛋白表达0.5 h开始增加(P<0.05),2 h达高峰(P<0.01),6 h开始下降,c-myc表达在24 h同对照组,但BDNF表达在24 h仍高于对照组。两者在不同区域、不同时间之间的表达不完全一致。结论BDNF和c-myc在弹道挫伤区、震荡区、海马及脑干神经元均有表达,两者在脑组织中表达的时空变化规律基本一致,可能共同参与细胞抗凋亡过程,相互间也可能存在着一定的调控作用。
Objective To study the changes of brain-derived neurotrophic factor (BDNF) and c-reactive protein c-myc after craniocerebral gunshot wound in dogs. Methods 24 mongrel dogs were randomly divided into control group (n = 4) and gunshot wound group (n = 20). Using German small-caliber rifle bullets canine cranial frontal tangential wound. Immunohistochemistry was used to detect the expression of BDNF and c-myc in the traumatic brain contusion, concussion zone, hippocampus and brainstem neurons at different time points of 0.5, 2, 6, 12 and 24 h after injury. Results The neuronal expression of BDNF and c-myc in the control group was weak. The expression of BDNF and c-myc protein in contusions, concussion zone, hippocampus and brain stem neurons in gunshot wound group increased 0.5 h (P <0.05), peaked at 2 h (P <0.01), decreased at 6 h, myc expression at 24 h with the control group, but BDNF expression was still higher than the control group at 24 h. The expression of the two in different regions and at different times is not exactly the same. CONCLUSION BDNF and c-myc are expressed in the ballistic contusions, concussion zone, hippocampus and brainstem neurons. The temporal and spatial changes of BDNF and c-myc in brain tissue are basically the same, which may be involved in the anti-apoptotic process of cells. There may be some regulation.
