目的研究肿瘤坏死因子α(TNF-α)诱导对膀胱癌细胞NF-κB通路分子表达及增殖水平的影响。方法TNF-α处理T24、5637膀胱癌细胞株,实时荧光定量PCR检测NF-κB信号通路Rel A、Rel B基因表达,CCK-8法检测细胞增殖活性。结果 T24细胞Rel A、Rel B表达在作用3 h和1 h后出现上调,5637细胞Rel A表达在TNF-α作用3 h后显著升高,差异均有统计学意义(P<0.05),Rel B表达差异无统计学意义(P>0.05)。TNF-α作用3 h的Rel A基因在2株细胞表达上调程度差异有统计学意义(P<0.05);TNF-α作用不同时间点的Rel B表达改变程度在2株细胞差异均有统计学意义(P<0.01)。TNF-α作用于2株细胞后,处理组和阴性对照组的存活率差异均无统计学意义(P>0.05)。结论膀胱癌细胞NF-κB信号通路Rel A、Rel B表达上调可能是TNF-α作用的早期事件,TNF-α对该通路的作用具有细胞选择性和不平衡性,其对膀胱癌细胞增殖活性无显著影响。
Objective To study the effect of tumor necrosis factor α (TNF-α) on the expression of NF-κB pathway and the proliferation of bladder cancer cells. Methods T24 and 5637 bladder cancer cell lines were treated with TNF-α. The expression of Rel A and Rel B genes of NF-κB signaling pathway was detected by real-time fluorescence quantitative PCR. The cell proliferation activity was detected by CCK-8 assay. Results The expression of Rel A and Rel B in T24 cells was up-regulated at 3 h and 1 h after treatment, and Rel A expression in 5637 cells was significantly increased 3 h after TNF-α treatment (P <0.05). Rel B expression difference was not statistically significant (P> 0.05). The upregulation of Rel A gene in 3 h after TNF-α treatment was statistically significant (P <0.05). The changes of Rel B expression at different time points after TNF-α treatment were statistically different between the two cell lines Significance (P <0.01). There was no significant difference in the survival rates between the two groups after treated with TNF-α (P> 0.05). Conclusion Up-regulation of NF-κB signaling pathway Rel A and Rel B in bladder cancer cells may be an early event of TNF-α. The effect of TNF-α on this pathway is cell-selective and unbalanced, and its effect on the proliferation of bladder cancer cells No significant effect.
