目的探讨人第10号染色体缺失性磷酸酶-张力蛋白同源物基因(PTEN)、基质金属蛋白酶-7(MMP-7)和血管内皮细胞生长因子(VEGF)在口腔鳞状细胞癌(OSCC)中的表达以及相关性。方法采用免疫组化SP法,分别检测40例OSCC组织和40例正常口腔黏膜组织中PTEN、MMP-7和VEGF的表达情况,并分析PTEN、MMP-7和VEGF在OSCC中表达的相关性。结果 PTEN、MMP-7和VEGF在OSCC组织中的表达率分别为35.0%(14/40)、65.0%(26/40)和72.5%(29/40),在正常口腔黏膜组织中的表达率分别为77.5%(31/40)、22.5%(9/40)和25.0%(10/40),3个指标在两种组织间的表达差异均有统计学意义(P<0.05)。OSCC组织中,PTEN表达与MMP-7及VEGF表达均呈负相关(r分别为-0.406和-0.394,均P<0.05);MMP-7表达与VEGF表达呈正相关(r=0.514,P<0.05)。结论 PTEN表达下调和MMP-7、VEGF表达上调与OSCC的发生发展密切相关;联合检测三者的表达对了解OSCC的恶性行为有重要的临床参考价值。
Objective To investigate the expression of PTEN, MMP-7 and VEGF in human oral squamous cell carcinoma (OSCC) In the expression and relevance. Methods Immunohistochemical SP method was used to detect the expression of PTEN, MMP-7 and VEGF in 40 OSCC tissues and 40 normal oral mucosa tissues, and to analyze the correlation between the expression of PTEN, MMP-7 and VEGF in OSCC. Results The positive rates of PTEN, MMP-7 and VEGF in OSCC tissues were 35.0% (14/40), 65.0% (26/40) and 72.5% (29/40), respectively. The positive rates of PTEN, 77.5% (31/40), 22.5% (9/40) and 25.0% (10/40) respectively. There was significant difference in the expression of the three indexes between the two tissues (P <0.05). In OSCC, the expression of PTEN was negatively correlated with the expression of MMP-7 and VEGF (r = -0.406 and -0.394, respectively, all P <0.05); the expression of MMP-7 was positively correlated with the expression of VEGF ). Conclusions The down-regulation of PTEN expression and the up-regulation of MMP-7 and VEGF expressions are closely related to the occurrence and development of OSCC. The combined detection of these three expressions has important clinical reference value for understanding the malignant behavior of OSCC.
