[目的] 观察铅对体外培养神经细胞静息状态45 Ca摄取及去极化后钙内流的影响。[方法] 培养的胎鼠神经细胞,通过铅和钙通道激动剂与对抗剂处理后,用β-液体闪烁计数器测量神经细胞中放射性45 Ca 的摄取速度。[结果] 低浓度铅对神经细胞45 Ca 摄取有抑制作用,但对神经细胞在静息时45Ca 摄取的速度无明显影响,去极化时抑制作用更加明显,并可分别被L型钙通道激动剂BayK8644所增强,对抗剂尼群地平所阻抑。[结论] 铅通过电压依赖性L-型钙通道对钙入胞产生抑制作用。
[Objective] To observe the effect of lead on 45 Ca uptake in resting nerve cells in vitro and calcium influx after depolarization. [Methods] The cultured fetal rat neurons were treated with lead and calcium channel agonists and antagonists, and then radioactive 45 Ca uptake rate in nerve cells was measured by β-scintillation counter. [Result] Low concentration of lead inhibited the uptake of 45 Ca in nerve cells, but had no obvious effect on the uptake rate of 45 Ca in resting neurons. The inhibition was more obvious when depolarized, and was activated by L - type calcium channels Agent BayK8644 enhanced, antagonist nitrendipine suppressed. [Conclusion] Lead can inhibit calcium entry through voltage-dependent L-type calcium channel.
