目的:观察宽心解郁汤、短期有氧运动及二者联合干预的抗抑郁作用,探讨可能机制。方法:SPF级雄性昆明小鼠,随机分成空白对照组、丙咪嗪阳性对照组、宽心解郁汤低剂量组(生药1.8 g/kg)、宽心解郁汤高剂量组(生药3.6 g/kg)、运动组(游泳30 min/d)、联合组(宽心解郁汤低剂量联合运动),干预8 d;通过强迫游泳、悬尾实验以及高架十字迷宫实验评价抗抑郁效果;处死小鼠,ELISA法检测脑组织单胺类神经递质含量,Western blotting分析脑组织BDNF表达。结果:宽心解郁汤高剂量表现出与丙咪嗪相似的抗抑郁效果,而低剂量抗抑郁效果不显著。单独的运动有与丙咪嗪相似或更好的抗抑郁效果。联合组疗效最佳。与空白对照组比较,宽心解郁汤高剂量组脑组织NE、DA含量以及运动组脑组织NE、DA及5-HT含量显著升高,丙咪嗪组、宽心解郁汤高剂量组、运动组及联合组脑组织BDNF蛋白表达显著高于空白对照组,运动组、联合组脑组织BDNF蛋白表达高于宽心解郁汤低、高剂量组,联合组脑组织BDNF蛋白显著升高。联合组脑组织NE、DA及5-HT含量显著高于阳性对照组。结论:适量宽心解郁汤、短期有氧运动以及联合干预对轻度抑郁有显著的抗抑郁疗效,其机制可能与增加脑组织NE、5-HT含量,引起脑组织BDNF蛋白表达增多,进而影响神经再生及可塑性、改善认知功能有关。

Methods: SPF male Kunming mice were randomly divided into 4 groups: blank control group, imipramine positive control group, low dose of widexinjieyu decoction (crude drug 1.8g / kg), high dose of widexinjieyu decoction (crude drug 3.6g / kg ), Exercise group (swimming 30 min / d), combination group (widexin Jieyu Decoction low-dose combination exercise) for 8 days; forced swimming, tail suspension test and elevated plus maze test to evaluate antidepressant effect; The content of monoamine neurotransmitter in brain tissue was detected by ELISA and the expression of BDNF in brain tissue was analyzed by Western blotting. Results: The wide dose of Jiexin Jieyu high dose showed imipramine similar antidepressant effect, and low dose antidepressant effect is not significant. Individual exercises have antidepressant effects similar to or better than imipramine. Combination group the best effect. Compared with the blank control group, the content of NE and DA in brain tissue of high-dose Xinxuetangyu Decoction group and the content of NE, DA and 5-HT in brain tissue in exercise group were significantly increased, while those in high-dose imipramine group, BDNF protein expression was significantly higher in brain tissue of combined group and control group than that in control group. BDNF protein in brain tissue of exercise group and combined group was higher than that of KHXT group, BDNF protein in brain tissue of combined group was significantly higher. The content of NE, DA and 5-HT in brain tissue of combined group was significantly higher than that of positive control group. CONCLUSION: Moderate relieving qi stagnation decoction, short-term aerobic exercise and combined intervention have significant antidepressant effect on mild depression. The mechanism may be related to increasing the content of NE and 5-HT in brain tissue, and increasing the expression of BDNF protein in brain tissue Nerve regeneration and plasticity, improve cognitive function.
