目的探讨少突胶质细胞转录因子(Olig)和神经钙黏附蛋白(N-cadherin)在人胚胎脊髓发育阶段的分布规律及其表达意义。方法应用免疫组织化学法检测第2、3、4月龄段,共17例人胚胎,脊髓中Olig和N-cadherin的表达和分布状况。应用One-Way ANOVA和LSD-t统计学方法,分析第2~4个月龄段人胚胎脊髓组织内N-cadherin和Olig蛋白的各自阳性表达强度值及Olig蛋白的阳性细胞表达数量。结果第2~4个月龄段,随着胎龄的增大,N-cadherin在脊髓组织的阳性表达强度值逐渐增大,其值分别是117.77±6.72、129.63±9.44和133.35±8.15,差异有统计学意义(P<0.01);Olig阳性表达强度值呈先升高再降低,其值分别是69.98±24.59、85.10±9.19、71.62±11.81,差异有统计学意义(P<0.05);Olig阳性表达细胞数量逐渐增多,其值分别是9.38±7.80、15.55±7.30、25.59±14.54,差异有统计学意义(P<0.01)。结论 N-cadherin和Olig在第2~4个月龄段人胚胎脊髓组织内均有阳性表达,提示其表达分布与脊髓组织内神经细胞和神经胶质细胞的分化、发育关系密切。
Objective To investigate the distribution and expression of Olig and N-cadherin in human embryonic spinal cord development. Methods Immunohistochemistry was used to detect the expression and distribution of Olig and N-cadherin in the second, third, and fourth month of age in 17 human embryos and spinal cord. One-Way ANOVA and LSD-t statistical methods were used to analyze the positive expressions of N-cadherin and Olig protein and the expression of Olig protein in human embryonic spinal cord tissues from the second to the fourth month. Results The positive expression of N-cadherin in the spinal cord tissue gradually increased with the increase of gestational age from the second to the fourth month of age, with the values of 117.77 ± 6.72, 129.63 ± 9.44 and 133.35 ± 8.15, respectively (P <0.01). The positive expression of Olig increased first and then decreased, with the values of 69.98 ± 24.59,85.10 ± 9.19 and 71.62 ± 11.81, respectively, with statistical significance (P <0.05); Olig The number of positive cells gradually increased, the values were 9.38 ± 7.80,15.55 ± 7.30,25.59 ± 14.54, the difference was statistically significant (P <0.01). Conclusions Both N-cadherin and Olig are positive in human embryonic spinal cord tissues from the second to the fourth month of age, suggesting that the expression and distribution of N-cadherin and Olig are closely related to the differentiation and development of nerve cells and glial cells in the spinal cord.
