目的探讨p130 cas在非小细胞肺癌中的表达与E-cadherin/β-catenin复合体的关系及对肿瘤细胞侵袭能力的影响。方法应用免疫组化方法检测p130 cas在145例非小细胞肺癌配对癌旁正常肺组织中的表达,并分析其表达与E-cadherin及β-catenin表达的相关性;同时进一步通过siRNA技术在肺癌细胞系中下调p130 cas的表达,观察E-cadherin和β-catenin表达的变化及肺癌细胞侵袭能力的变化。结果 p130 cas在非小细胞肺癌组织标本中表达上调(P<0.001),并与E-cadherin和β-catenin的表达下调相关(P=0.001和P=0.003);Western blot结果显示在肺癌细胞系A549和H1299中下调p130 cas的表达可以使E-cadherin和β-catenin的表达上调,而且侵袭实验结果显示下调p130 cas的表达可以使肺癌细胞的侵袭能力下降。结论 p130 cas在肺癌中可能通过下调E-cadherin/β-catenin复合体的表达促进肿瘤细胞的侵袭和转移。
Objective To investigate the relationship between the expression of p130 cas in non-small cell lung cancer (NSCLC) and the E-cadherin / β-catenin complex and its effect on tumor cell invasiveness. Methods Immunohistochemistry was used to detect the expression of p130 cas in 145 non-small cell lung cancer matched normal lung tissues. The correlation between the expression of E-cadherin and β-catenin was analyzed. At the same time, The expression of p130 cas was down-regulated in cell lines and the expression of E-cadherin and β-catenin and the invasion ability of lung cancer cells were observed. Results The expression of p130 cas was up-regulated in non-small cell lung cancer (P <0.001) and correlated with the down-regulation of E-cadherin and β-catenin (P = 0.001 and P = 0.003) Down-regulation of p130 caspase expression in A549 and H1299 up-regulated the expression of E-cadherin and β-catenin, and the invasion assay showed that the down-regulation of p130 cas expression could decrease the invasion ability of lung cancer cells. Conclusion p130 cas may promote tumor cell invasion and metastasis by down-regulating the expression of E-cadherin / β-catenin complex in lung cancer.
