CD8 ̄+T细胞识别的抗原为靶细胞或抗原递呈细胞MHCⅠ类分子递呈的多肽,它是抗原性物质经过细胞内加工后并由MHCⅠ类分子从细胞内递呈出来。利用酸洗法获得MHCⅠ类分子递呈的多肽,然后对其进行活性鉴定及顺序分析,采用合成肽进行对比的功能检测,发现MHCⅠ类分子递呈的多肽为8~10个氨基酸组成,结构中含有保守性氨基酸;同一MHC等位型具有一致的MHC特异的多肽结合关键序列(MHChindingmotif),从而为合成抗原性多肽提供了重要依据。
Antigens recognized by CD8 + T cells are peptides presented by MHC class I molecules of target cells or antigen-presenting cells, which are produced by intracellular processing of antigenic material and by MHC class I molecules. The MHC class I molecules were obtained by pickling method, and then their activity was identified and sequenced. Using the synthetic peptides for functional comparison, the peptides presented by MHC class I molecules were composed of 8 to 10 amino acids. In the structure Contains a conserved amino acid; the same MHC allele has a consistent MHC-specific peptide binding key sequence (MHChindingmotif), which provides an important basis for the synthesis of antigenic polypeptides.
