建立了利用电场轨道阱回旋共振组合质谱仪(LTQ-Orbitrap)检测人体尿样中青霉素G及其代谢产物的分析方法。目标化合物分离鉴定选用PLRP-S聚合色谱柱,以含0.1%(体积比)甲酸溶液-乙腈为流动相,梯度洗脱分离后,选用Data-dependent MSn模式进行全扫描分析。尿样中首次检出青霉素甲酰化代谢产物(m/z 381)。青霉素新代谢物的发现对食品安全及法医毒物分析具有重要意义。
A method for the determination of penicillin G and its metabolites in human urine samples by using the electric field orbit trapper cyclotron resonance mass spectrometer (LTQ-Orbitrap) was established. The target compound was isolated and identified by PLRP-S polymerization column. The mobile phase consisted of 0.1% (volume) formic acid solution-acetonitrile as the mobile phase. After gradient elution, the data were analyzed by Data-dependent MSn mode. Penicillin formylation metabolites (m / z 381) were first detected in urine samples. The discovery of new metabolites of penicillin is of great importance for food safety and forensic toxicological analysis.
