目的 :研究纤维蛋白胶对体外培养小鼠胚胎发育的影响。方法 :超排获取昆明小鼠 2细胞期受精卵 ,常规配制纤维蛋白胶 ,比较小鼠胚胎常规培养与在纤维蛋白胶上培养的囊胚发育率及孵化率 ,以及内细胞团染色体数目变化。结果 :小鼠胚胎在纤维蛋白胶上培养与常规培养 ,囊胚发育率及孵化率分别为 :6 7.5 %和 6 6 .7% (P =0 .837,Yetescorrected) ,38.5 %和 34.0% (P =0 .15 9,Yetescorrected)。内细胞团染色体数目绝大部分为 2n =4 0 ,基本保持核型稳定。结论 :纤维蛋白胶对体外培养的小鼠胚胎无不良影响 ,可以作为研究胚胎营养因子对胚胎影响的载体基质。
Objective: To study the effect of fibrin glue on embryonic development in vitro. Methods: Two-cell fertilized eggs of Kunming mice were obtained by superovulation. Fibrin glue was prepared routinely. The developmental and hatching rates of blastocysts cultured in the conventional culture of mouse embryos and on fibrin glue were compared with those of the inner cell mass. Results: The mouse embryos were cultured on fibrin glue and cultured routinely. The blastocyst development rate and hatching rate were 6 7.5% and 66.7% (P = 0.387, Yetescorrected), 38.5% and 34.0% respectively P = 0 .15 9, Yetescorrected). Most of the inner cell mass chromosome number 2n = 40, the basic stable karyotype. CONCLUSION: Fibrin glue has no adverse effect on mouse embryos cultured in vitro and can be used as carrier matrix to study the effect of embryonic trophic factors on embryos.
