目的:体外实验观察绞股蓝、黄芩甙、丹参酚酸B、灯盏花乙素4种中草药对人颊癌细胞株(HIOEC-BaP,HB)的抑制作用,选取其中抑制效果最佳的两种中药观察其对HB的抑制作用。方法:采用四唑盐(PMS-MTT)比色法检测绞股蓝、黄芩甙、丹参酚酸B、灯盏花乙素4种中草药对HB的抑制作用,并与西药(顺铂)做对比,计算出各药梯度剂量的细胞存活百分率,绘制药效曲线图。采用中效原理计算出各药的ID50,选出其中抑制效果最佳的两种中药复方合用,观察其对HB的抑制作用,计算出梯度剂量的细胞存活百分率,绘制药效曲线图。采用中效原理计算出ID50,判断两种药物的相互作用关系。结果:绞股蓝、黄芩甙、丹参酚酸B、灯盏花乙素4种中草药对人颊癌细胞株(HB)有抑制作用,经方差分析,4种中药与顺铂对人颊癌细胞株(HB)的抑制作用有统计学差异﹙P<0.01),而4种中药之间HB的抑制作用也有统计学差异﹙P<0.01),其中黄芩甙、灯盏花乙素的抑制作用最佳。它们的ID50分别是黄芩甙:391.7832μM;灯盏花乙素:354.6062μM。黄芩甙、灯盏花乙素对这种细胞株的抑制率的差异无统计学意义(P>0.05)。中药黄芩甙、灯盏花乙素合用对HB的ID50是208.222μM,这两种中药的合用指数CI<1,表明有协同作用。结论:绞股蓝、黄芩甙、丹参酚酸B、灯盏花乙素4种中草药对HB细胞株有一定的抑制作用,其中黄芩甙、灯盏花乙素对细胞株的抑制作用最佳。且两种中药复方时对人颊癌细胞株有协同抑制作用。
OBJECTIVE: To observe the inhibitory effects of four herbs of Gynostemma pentaphyllum, Astragalus membranaceus, salvianolic acid B, and scutellarin on human cheil cancer cell lines (HIOEC-BaP, HB) in vitro and select the two herbs with the best inhibitory effect Its inhibitory effect on HB. Methods: The inhibitory effect of four herbs (Gynostemma pentaphyllum, Astragalus membranaceus, salvianolic acid B, scutellarin) on HB was detected by tetrazolium salt (PMS-MTT) colorimetric method and compared with western medicine (cisplatin). Results: Gynostemma pentaphyllum, Astragalus membranaceus, Salvia miltiorrhiza B, and scutellarin inhibited human cheek cancer cell line (HB). After analysis of variance, four Chinese herbs and cisplatin against human cheek cancer cell line (HB There was a statistically significant difference (P<0.01) in the inhibition of HBsAg, and there was a statistically significant difference in the inhibitory effect of HB between the four herbs (P<0.01). Among them, Astragalus membranaceus and Erigeron breviscapinis had the best inhibitory effect. Their ID50s are respectively Astragalus: 391.7832 μM; scutellarin: 354.6062 μM. There was no significant difference in the inhibition rate of Astragalus and scutellarin on this cell line (P>0.05). The ID50 of HBsAg combined with Radix Astragali and Erigeron breviscapus was 208.222 μM. The combined index CI of these two herbs was CI<1, indicating a synergistic effect. Conclusion: Gynostemma pentaphyllum, Astragalus membranaceus, salvianolic acid B and scutellarin can inhibit the HB cell line. Among them, Scutellaria baicalensis and scutellarin can inhibit the cell line. In addition, there are synergistic inhibitory effects on the human cheek cancer cell lines when the two Chinese herbal compounds are combined.
