目的研究资木瓜皂苷(SCS)提取工艺的优化及对大鼠佐剂性关节炎(AA)的抗炎作用及免疫机制。方法以SCS的提取总量为考察指标,采取正交试验法考察乙醇浓度、溶媒用量、提取时间、提取次数等因素,每个因素设置3个水平。流式细胞术检测SCS对AA外周血中CD4+、CD8+的表达水平;MTT法检测SCS对AA外周T淋巴细胞增殖的影响。结果 SCS最佳提取工艺为乙醇浓度70%,溶媒容量10倍,提取时间1h,提取3次,提取率为1.7%。SCS能降低AA外周血中CD4+的表达水平,对CD8+的表达无明显影响,对AA外周T淋巴细胞增殖有抑制作用。结论最佳提取条件为A2B1C2D3,SCS通过抑制T淋巴细胞增殖和降低CD4+表达而发挥抗炎免疫作用。
Objective To study the optimization of the extraction process of papaya saponin (SCS) and its anti-inflammatory effects and immune mechanism on adjuvant arthritis (AA) in rats. Methods Taking the total amount of SCS extracted as the inspection index, the orthogonal test method was used to examine the factors such as ethanol concentration, amount of solvent, extraction time, and number of extractions. Each factor was set at three levels. Flow cytometry was used to detect the expression of CD4+ and CD8+ in peripheral blood of AA. The effect of SCS on the proliferation of AA peripheral T lymphocytes was detected by MTT assay. Results The best extraction process of SCS was ethanol concentration 70%, solvent capacity 10 times, extraction time 1 h, extraction 3 times, extraction rate 1.7%. SCS can reduce the expression of CD4+ in peripheral blood of AA, and has no obvious effect on the expression of CD8+. It can inhibit the proliferation of AA peripheral T lymphocytes. Conclusion The optimal extraction condition is A2B1C2D3. SCS exerts anti-inflammatory effects by inhibiting the proliferation of T lymphocytes and decreasing the expression of CD4+.
