建立人中性粒细胞（PMN）防御素（HNP）的制备级纯化方法，以便更深入研究防御素的生物学活性．方法：采用Superdex30凝胶色谱法对PMN提取物进行初步分离，再用C18反相液相色谱法进一步纯化，得到HNP纯品并对其进行鉴定．结果：纯化的HNPMr为3400，纯度为91．6％，具有抗微生物活性．结论：应用两步色谱法纯化HNP，是从人的PMN中纯化HNP的较为理想的方法．
A preparative purification method of human neutrophil (PMN) defensin (HNP) was established to further investigate the biological activity of defensins. Methods: PMN extracts were separated by Superdex30 gel chromatography and further purified by C18 reversed-phase liquid chromatography to obtain pure HNP samples. Results: The purified HNPMr was 3400 with 91.6% purity and had antimicrobial activity. Conclusion: HNP purification by two-step chromatography is an ideal method to purify HNP from human PMN.
