本文用高效液相色谱法测定人血浆中布洛芬的浓度,以消炎痛为内标,用正已烷:氯仿:异丙醇(65:25:10)混合溶剂提取,以Zorbax—C_8柱作固定相,以甲醇:0.1mol/L醋酸缓冲液(pH=4.7):0.085%的磷酸溶液(135:35:30)为流动相,225nm为检测波长,用峰高比定量。线性范围为0.5~40μg/ml,最低检测浓度为0.1μg/ml.用本法测定了健康人口服布洛芬片后的血药浓度,进行了药代动力学参数计算。
In this paper, the concentration of ibuprofen in human plasma was determined by HPLC. Indomethacin was used as the internal standard and extracted with a mixed solvent of n-hexane: chloroform: isopropanol (65:25:10). The column was eluted with Zorbax-C_8 The stationary phase was eluted with a mobile phase of methanol: 0.1mol / L acetate buffer (pH = 4.7): 0.085% phosphoric acid solution (135: 35: 30) at a detection wavelength of 225nm. The linear range was 0.5 ~ 40μg / ml, the lowest detection concentration was 0.1μg / ml.This method was used to determine the blood concentration of healthy people after oral administration of ibuprofen tablets, and the pharmacokinetic parameters were calculated.
