PROL4(proline-rich protein 4)是我们筛选发现的一个在肺癌细胞中表达缺失的基因.为了深入研究其在肺癌中的生物学功能,构建了PROL4表达质粒,转染肺腺癌LTEP-a-2细胞,获得了稳定表达PROL4的单克隆细胞株.与空载对照细胞相比,过表达PROL4的LTEP-a-2细胞其生长的血清依赖性明显提高,软琼脂集落形成能力降低,裸鼠皮下荷瘤生长受到明显抑制.进一步研究发现,过表达PROL4可明显提高多种肺癌细胞对顺铂的敏感性,而且这种敏感性的提高与PROL4协同顺铂促进细胞凋亡密切相关.该研究结果为临床上如何提高肺癌的化疗效果提供了重要的实验依据.
PROL4 (proline-rich protein 4) is one of the genes that we found in lung cancer cells.Expression of PROL4 gene was cloned in order to further study its biological function in lung cancer. PROL4 expression plasmid was constructed and transfected into lung adenocarcinoma LTEP-a- 2 cells, a monoclonal cell strain stably expressing PROL4 was obtained.Protective growth of LTEP-a-2 cells overexpressing PROL4 was significantly increased compared with that of the control cells, the soft agar colony-forming ability was reduced, and nude mice Subcutaneous tumor growth was significantly inhibited.Further study found that over-expression of PROL4 can significantly improve the sensitivity of a variety of lung cancer cells to cisplatin, and this increase in sensitivity and PROL4 synergistic with cisplatin to promote apoptosis is closely related to the study The results provide an important experimental basis for how to improve the chemotherapy effect of lung cancer clinically.
