目的 :观察辛伐他汀对骨髓基质细胞成骨分化的影响 ,探讨其刺激成骨的作用机制。方法 :体外培养成年小鼠骨髓基质细胞 ,不同浓度的辛伐他汀作用 72h后 ,RT PCR检测骨钙素 (osteocalcin ,OCN)mRNA水平的变化 ,Westernblot检测OCN和骨桥蛋白 (osteopontin ,OPN)表达水平的变化 ,细胞碱性磷酸酶 (alkalinephosphatase,ALP)细胞化学染色及酶比活性测定。结果 :辛伐他汀作用 72h后 ,OCN的mRNA表达水平增高 ,OCN、OPN蛋白表达水平增高 ,呈剂量依赖关系 ,细胞ALP染色增强 ,ALP比活性显著增高。结论 :辛伐他汀促进骨髓基质细胞的成骨分化 ,辛伐他汀刺激成骨的作用机制可能与此有关
OBJECTIVE: To observe the effect of simvastatin on osteogenic differentiation of bone marrow stromal cells (BMSCs) and to explore its mechanism of stimulating osteogenesis. Methods: Adult mouse bone marrow stromal cells were cultured in vitro. The mRNA expression of osteocalcin (OCN) was detected by RT-PCR after 72 h treatment with different concentrations of simvastatin. The expressions of OCN and osteopontin (OPN) were detected by Western blot Level changes, cytochemical staining of alkaline phosphatase (ALP) and enzyme activity determination. RESULTS: After 72 hours of simvastatin treatment, OCN mRNA expression increased, OCN and OPN protein expression increased in a dose-dependent manner, ALP staining increased and ALP specific activity increased significantly. Conclusion: Simvastatin can promote osteogenic differentiation of bone marrow stromal cells and the mechanism of simvastatin stimulating osteogenesis may be related to this
