目的:探讨丹参酮IIA和振动运动对2型糖尿病大鼠肝脏损伤的保护作用及其机制。方法:腹腔注射链脲佐菌素和高脂饮食相结合的方法建立大鼠2型糖尿病模型。实验分为正常组、模型组、丹参酮IIA(10 mg/kg)组、丹参酮IIA(20 mg/kg)组、振动运动组、丹参酮IIA(10 mg/kg)+振动运动组、丹参酮IIA(20 mg/kg)+振动运动组,尾静脉采血测定大鼠血糖水平,ELESA法测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性和超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量;HE染色观察肝组织细胞结构变化;Western blot法测定肝组织核因子κB(NF-κB)p65蛋白表达水平。结果:模型组空腹血糖与正常组相比显著增加,血清ALT、AST活性增加,TNF-α、IL-6和MDA含量均增加,SOD活力降低,NF-κBp65表达明显增加。丹参酮IIA(20 mg/kg)+振动运动组空腹血糖水平与模型组相比显著降低,血清ALT和AST活性降低,TNF-α、IL-6和MDA含量均降低,SOD活力升高,而NF-κBp65在肝组织核蛋白中的表达降低。结论:丹参酮IIA联合振动运动对糖尿病肝损伤具有一定的保护作用,这种保护作用与抑制氧化应激和炎症反应有关。
Objective: To investigate the protective effects of tanshinone IIA and vibrational exercise on liver injury in type 2 diabetic rats and its mechanism. Methods: A rat model of type 2 diabetes was established by intraperitoneal injection of streptozotocin and high fat diet. CONCLUSION: Tanshinone IIA combined with vibrational exercise has a protective effect on diabetic liver injury. This protective effect is related to inhibition of oxidative stress and inflammatory response.
